Sucessivos escândalos de manipulação de demonstrações financeiras das companhias abertas nos Estados Unidos da América, nos anos recentes, motivaram suas autoridades reguladoras a promulgar a Lei Sarbanes-Oxley (Sox). Essa estabeleceu um conjunto severo de regras às principais figuras envolvidas no mundo corporativo, abarcando administradores, auditores, advogados e analistas de mercado. Aos auditores independentes foram destinadas regras específicas que introduziram novidades como a criação de órgão fiscalizador da profissão (PCAOB), novas regras de independência e renovadas normas de auditoria (auditing standards). A pesquisa cobriu o objetivo principal de analisar os impactos (variáveis dependentes) percebidos pelos auditores independentes com a introdução da lei estadunidense (variável independente), utilizando como ferramenta a aplicação de um questionário. A metodologia de pesquisa combina revisão bibliográfica do tema e levantamento da percepção dos impactos da Sox pelos auditores independentes. Para os pesquisados, a Sox aumentou a fiscalização sobre a profissão; agregou maior credibilidade às demonstrações financeiras, ao trabalho de auditores e contadores e contribuiu para o aumento dos honorários de auditoria.

Successive scandals of manipulation of open corporations' financial statements in the United States, in recent years, motivated their regulatory authorities to enact the Sarbanes-Oxley Act ("Sox"). This established a strict set of rules for the primary figures involved in the corporate world, including managers, auditors, lawyers and market analysts. Specific rules that introduced innovations, such as the creation of a supervisory body (PCAOB), new independence rules and renewed auditing standards were designed for independent auditors. The research covered the main goal of analyzing the impacts (dependent variables) independent auditors perceived after the introduction of the American law (independent variable), through a questionnaire. The research methodology combines a bibliographic review and a survey of how independent auditors perceived the Sox Act impacts. According to the participants, the Sox Act increased the supervision on the profession; added more credibility to the financial statements, to the work of auditors and accountants, and contributed to the raising of audit fees.

No presente trabalho, foi purificada por cromatografia de afinidade em D-galactose imobilizada em agarose, uma lectina do latex de Synadenium carinatum (ScLL). Essa lectina é uma potente aglutinina para eritrócitos humanos, cuja atividade hemaglutinante foi inibida com 3,0 mM de N acetil-D-galactopiranosidio, 6,3 mM de metil-beta-D-galactopiranosidio ou 50 mM metil-alfa-D-pironosidio ou D-fucose, porém, nenhuma inibição foi evidenciada por L-fucose, revelando uma especificidade anomérica e conformacional da lectina. A análise por SDS-PAGE dessa lectina pareceu ser uma glicoproteína composta por duas cadeias polipeptídicas de aproximadamente 28 e 30 kDa, porém, em cromatografia de exclusão por tamanho sobre Sephadex G100 e em gel nativo apresentou um peso molecular aparente de 120-130 kDa, a qual mostrou ser composta de uma mistura de subunidades de peptídeos de 28 e 30 kDa. Essa lectina manteve-se estável em pH de 6 a 9 e temperatura de 4 a 56ºC. A seqüência N-terninal contem uma região conservada GPN a qual também é observada em outras lectinas de látex de outras Euphorbiaceas ligante de D-galactose.

A lectin from the latex of Synadenium carinatum was purified by affinity chromatography on immobilized-D-galactose-agarose and shown to be a potent agglutinin of human erythrocytes. The haemagglutination of human red cells was inhibited by 3.0 mM N-acetyl-D-galactopyranoside, 6.3 mM methyl-beta-D-galactopyranoside, 50 mM methyl-alpha-D-galactopyranoside and 50 mM D-fucose but not by L-fucose, demonstrating an anomeric and a conformational specificity. According to SDS-PAGE analysis, the lectin appeared to be a glycoprotein composed of two polypeptide chains of ca. 28 and 30 kDa, but size exclusion chromatography (Sephadex G-100) and native PAGE revealed a protein of apparent molecular weight 120 - 130 kDa made up of 28 and 30 kDa subunits. The lectin was stable in the range pH 6 - 9, and 4 - 56ºC. The N-terminal sequence of the 30 kDa subunit contained the conserved consensus sequence GPN observed in other D-galactose-binding lectins found in latex of members of the Euphorbiaceae.

O presente trabalho avaliou o perfil de anticorpos em amostras de soro de 37 pacientes com diagnóstico clínico confirmado ou compatível com leishmaniose tegumentar americana atendidos no Hospital de Clínicas da Universidade Federal de Uberlândia, MG. Os perfis das classes de imunoglobulinas e subclasses de IgG foram analisados pelo teste ELISA indireto, utilizando-se antígeno solúvel de Leishmania (Leishmania) amazonensis. A avidez dos anticorpos foi determinada pelo tratamento com uréia a 6 M, após incubação dos soros com o antígeno. Observou-se que 97%, 94,6%, 57,5 e 21,5% das amostras testadas apresentaram anticorpos anti-Leishmania das classes IgE, IgG, IgA e IgM, respectivamente e, os perfis das subclasses de IgG demonstraram, IgG1>IgG3>IgG2>IgG4. Os anticorpos IgE anti-Leishmania de alta avidez corresponderam a 44,4%. Por outro lado, IgG e IgA anti-Leishmania foram em sua maioria (62,8 e 47,8%, respectivamente), de média avidez. A variação do perfil de isotipos, bem como a avidez das imunoglobulinas refletiu a complexidade da resposta imune humoral contra a leishmaniose tegumentar americana.

The present work investigated the serum antibody profiles in 37 patients with American tegumentary leishmaniasis, who were attended at Hospital de Clinicas - Universidade Federal de Uberlandia, MG, Brazil. The immunoglobulin class and IgG subclass profiles were analyzed by indirect ELISA using Leishmania (Leishmania) amazonensis soluble antigen. The antibody avidity was determined by 6 M urea treatment after incubation with immunoenzymatic conjugate. It was observed that 97% of the serum samples presented anti-Leishmania antibodies for IgE class, 94.6% IgG, 57.5% IgA and 21.5% IgM class. For IgG subclasses the profiles were in the following order of frequency: IgG1>IgG3>IgG2>IgG4. High avidity of anti-Leishmania IgE antibodies was found in 44.4% of the samples. On the other hand, moderate avidity of specific IgG and IgA was observed in 62.8% and 47.8% of samples, respectively. These results indicate a very complex antibody response profile against American tegumentary leishmaniasis.
